Tobi Amusan began her athletic career by earning the silver medal at the African Youth Championships in Warri in 2013 and the gold medal in the 100-meter hurdles at the African Junior Athletics Championships in Addis Abeba in 2015. In 2015, when she was eighteen years old and competing in her first All-Africa Games, she took home the gold in the 100-meter hurdles. After winning gold in the 100 mH and 200 m, she became the second athlete for the school to be named C-USA Female Track Athlete of the Year since UTEP joined C-USA. She ran a timing of 12.83 seconds to become the first hurdler to break the 13-second barrier, and she finished second in the 100-meter hurdles at the 2016 NCAA Outdoor Championships.

Additionally, she participated in the 2016 World Junior Championships in Bydgoszcz before representing Nigeria at the 2016 Olympic Games in Rio, where she advanced to the 100 mH semifinals. Later in 2022, Tobi participated in a Diamond League competition and came in second place to Olympic champion Jasmine Camacho-Quinn with a time of 12.60 seconds. She also entered the 2022 World Championships as a medal possibility following consecutive fourth-place finishes in which she set a new African record of 12.40 seconds, beating her previous mark by 0.01 seconds. She became the first Nigerian global champion at the World Athletics Championships by setting a new world record of 12.12 seconds in the semifinal and breaking it again in the final with a time of 12.06 seconds.
