The Chicago Bulls selected Aldridge second overall in the 2006 NBA draft, but his rights were quickly dealt to the Portland Trail Blazers in exchange for their pick, Tyrus Thomas, and Viktor Khryapa. In an 87-95 loss against the Atlanta Hawks in 2012, Aldridge set a career high with eight assists. For the second time in his career, Aldridge was chosen an All-Star in 2013.

In 2015, he signed a four-year, $80 million deal with the San Antonio Spurs. Despite retiring for Bruce Bowen, Aldridge was given the number 12 jersey when he was purchased by the Spurs. In 2017, the Spurs defeated the Houston Rockets 114–75 in Game 6 of their second-round playoff series, with Aldridge scoring a season-high 34 points.

He also became the first Spur since Tim Duncan against the Phoenix Suns in 2008 to score 34 points and grab 12 rebounds in a playoff game.
