Pondexter was chosen 26th overall by the Oklahoma City Thunder in the 2010 NBA Draft. After a month, the New Orleans Hornets acquired Pondexter and Craig Brackins.
Before the start of the 2011-12 NBA season, the Hornets traded him to the Memphis Grizzlies. After two seasons with the Grizzlies, he signed a four-year, $14 million contract with the team in 2013. Pondexter appeared in 189 games in his first three NBA seasons.
However, he suffered a right leg injury during the 2013-14 NBA season and was forced to miss the rest of the season after undergoing surgery in December. Pondexter returned for the 2014-15 season, spending the first two months with the Grizzlies.
After that, he was traded back to New Orleans, now known as the Pelicans, in a three-team deal involving the Grizzlies and the Boston Celtics. He averaged 9 points per game and set a career high with 25 against the Brooklyn Nets.
Due to a series of injuries, he was forced to retire from the game, but he returned after a series of surgeries. He was also traded to the Chicago Bulls in 2017 and played against the Toronto Raptors while suffering from a left hamstring injury. He was let go by the Bulls.
