Sean Kenniff was born in the town of Massapequa, New York. He was born on November 27, 1969. His father worked for the city of New York as a firefighter, while his mother stayed at home.
Sean graduated from Massapequa High School and began his studies at the State University of New York at Binghamton in the fall of the following year, earning a Bachelor of Science in biology.

Sean stayed behind during spring break while his classmates went to college to save money for medical school. He received his M.D. from New York Medical College. After graduation, he worked for four years at the Long Island Jewish Medical Center, the last year as Chief Resident.
