He made his professional debut for the reserves side, Jong Ajax on 13 March 2015 in a 1-0 loss against VVV Venlo in an Eerste Divisie. Also, He then made his official first-team debut and also scored his first goal for the club on 21 September 2016 in a 5-0 win against Willem II. On 8 July 2017, at age 20, during a pre-season friendly match against Werder Bremen, he fell and suffered a cardiac arrhythmia event, which left him with severe and permanent brain damage and unable to continue his career as a player. In June 2018, Ajax announced an investigation had found that the medical treatment he received on the field was “inadequate”, which prompted a lawsuit from his family against Ajax through the Royal Dutch Football Association’s arbitration panel, who stated a failure to resuscitate him with due haste was responsible for causing his brain damage.
On 27 March 2020, his family stated that he had awoken from his coma, returned home to his family, and that they were caring for him. Shortly afterward, Ajax said they had canceled his contract, which was set to automatically renew on 1 July, and that they were in negotiations with his family about a “future solution” for him. On 21 February 2022, Ajax announce that they have came to an arrangement with his family that they will pay his family €7.8M owing to the insufficient treatment he had received after his fall on the pitch and they also have told the family that number 34 will get retired forever.

Following his retirement, various tributes were conducted in his honour by Ajax and players from other teams. In December 2018, Ajax renamed their “Talent of the Future” award for him, while a number of Nouri’s former Ajax and Dutch teammates, including Justin Kluivert, Philippe Sandler, Amin Younes, Kevin Diks, Joel Veltman, Donny van de Beek, Sofyan Amrabat, and Anwar EL Ghazi have adopted his shirt number (34) at their new clubs.
